The cells were precipitated with 5% ice-cold trichloroacetic acid. Precipitated pellets were washed with 80% cold acetone and then suspended in SDS sample buffer. SDS-polyacrylamide gel electrophoresis and Western blotting analysis were carried out as described above. The activity of β-galactosidase was assayed using o-nitrophenyl-β-d-galactopyranoside as the substrate. The specific activity was recorded as μmol of o-nitrophenol min−1 (mg cellular protein)−1 (Miller, 1972).
